Induction of lethal graft versus host disease in rats by spleen grafting

      Abstract

      Spleen allografts were exchanged between AGUS and PVG rats which have a strong Ag-B difference. Fatal graft versus host (GVH) reactions occurred in four donor/recipient combinations when:
      • 1.
        (1) AGUS spleens were grafted to (PVG × AGUS) F1 rats;
      • 2.
        (2) PVG spleens were grafted to sublethally irradiated AGUS recipients;
      • 3.
        (3) PVG spleens were grafted to AGUS rats, the donors being actively presensitized with recipient thymus cells and spleen cells;
      • 4.
        (4) PVG spleens were grafted to AGUS rats which had their own spleens removed on the day of operation.
      These findings point to the significance of the immunocompetence of the allografted spleen in this model. The fact that the recipient rats were not able to reject spleen grafts opens a field for further experiments. Controlled GVH might turn out to be a means of achieving spleen graft acceptance without the recipients succumbing to GVH.
